A weekend without electricity was already trying for millions in the sweltering, storm-swept mid-Atlantic region. But Monday morning brings another grim challenge when many embark on a difficult commute over roads with darkened stoplights and likely mass-transit delays.

To alleviate congestion around Baltimore and Washington, federal and state officials gave many workers the option of staying home Monday. Federal agencies will be open in Washington, but non-emergency employees have the option of taking leave or working from home. Maryland's governor also gave state workers wide leeway for staying out of the office.

As of Monday morning, around 2 million customers across a swath of states along the East Coast and as far west as Illinois remained without power. That left many to contend with stifling homes and spoiled food over the weekend as temperatures approached or exceeded 100 degrees, and utility officials said the power will likely be out for several more days. Since Friday, severe weather has been blamed for at least 17 deaths, most from trees falling on homes and cars.

There were more than 400 signal outages in Maryland on Monday, including more than 330 in hard-hit Montgomery County outside the nation's capital, according to the State Highway Administration. There were 100 signal outages in northern Virginia late Sunday afternoon, and 65 roads were closed, although most were secondary roads.

"If you have to drive or need to drive, leave yourself a lot of extra time," Maryland State Highway Administration spokesman Charlie Gischlar said. "There's going to be delays."

All 86 Metro subway stations in the Washington area were open, but delays were possible Monday because power was being routed through the system to serve some areas where power was not being supplied by commercial utilities, spokesman Dan Stessel said. Some stations in Montgomery County were running on backup power, he said, meaning escalators may not work ? bad news for commuters braving the stifling heat. Metro bus riders were expected to experience significant delays.

Drivers resorted to ingenuity to get to work. On a residential street in suburban Falls Church, Va., just outside Washington, downed trees blocked the road on either side. Enterprising neighbors used chain saws to cut a makeshift path on one side, but the other remained completely blocked by a massive oak tree.

"They kind of forgot about us out here," resident Eric Nesson said.

Still, residents took the aggravation with good humor. Posted on the oak tree was a sign saying: "Free firewood you haul." The tree lay across a smashed Ford pickup truck, with a sign reading: "For SALE. Recently lowered."

Meanwhile, Coast Guard officials say they have suspended the search for a man who disappeared early Saturday while boating during the storm off Maryland.

Researchers block pathway to cancer cell replication

ScienceDaily (July 2, 2012) ? Research suggests that patients with leukemia sometimes relapse because standard chemotherapy fails to kill the self-renewing leukemia initiating cells, often referred to as cancer stem cells. In such cancers, the cells lie dormant for a time, only to later begin cloning, resulting in a return and metastasis of the disease.

One such type of cancer is called pediatric T cell acute lymphoblastic leukemia, or T-ALL, often found in children, who have few treatment options beyond chemotherapy.

A team of researchers -- led by Catriona H. M. Jamieson, MD, PhD, associate professor of medicine at the University of California, San Diego School of Medicine and Director of Stem Cell Research at UC San Diego Moores Cancer Center -- studied these cells in mouse models that had been transplanted with human leukemia cells. They discovered that the leukemia initiating cells which clone, or replicate, themselves most robustly activate the NOTCH1 pathway, usually in the context of a mutation.

Earlier studies showed that as many as half of patients with T-ALL have mutations in the NOTCH1 pathway -- an evolutionarily conserved developmental pathway used during differentiation of many cell and tissue types. The new study shows that when NOTCH1 activation was inhibited in animal models using a monoclonal antibody, the leukemia initiating cells did not survive. In addition, the antibody treatment significantly reduced a subset of these cancer stem cells (identified by the presence of specific markers, CD2 and CD7, on the cell surface.)

"We were able to substantially reduce the potential of these cancer stem cells to self-renew," said Jamieson. "So we're not just getting rid of cancerous cells: we're getting to the root of their resistance to treatment -- leukemic stem cells that lie dormant."

The study results suggest that such therapy would also be effective in other types of cancer stem cells, such as those that cause breast cancer, that also rely on NOTCH1 for self-renewal.

"Therapies based on monoclonal antibodies that inhibit NOTCH 1 are much more selective than using gamma-secretase inhibitors, which also block other essential cellular functions in addition to the NOTCH1 signaling pathway," said contributor A. Thomas Look, MD of Dana-Farber/Children Hospital Cancer Center in Boston. "We are excited about the promise of NOTCH1-specific antibodies to counter resistance to therapy in T-ALL and possibly additional types of cancer."

In investigating the role of NOTCH1 activation in cancer cell cloning, the researchers showed that leukemia initiating cells possess enhanced survival and self-renewal potential in specific blood-cell, or hematopoietic, niches: the microenvironment of the body in which the cells live and self-renew.

The scientists studied the molecular characterization of CD34+ cells -- a protein that shows expression in early hematopoietic cells and that facilitates cell migration -- from a dozen T-ALL patient samples.

They found that mutations in NOTCH1 and other genes capable of promoting the survival of cancer stem cells co-existed in the CD34+ niche. Mice transplanted with CD34-enriched NOTCH1 mutated T-ALL cells demonstrated significantly greater leukemic cloning potential than did mice without the NOTCH1 mutation. The mutated cells were uniquely susceptible to targeted inhibition with a human monoclonal antibody, according to the scientists.

Some brains may be primed for pain

Addiction-related process may keep people hurting long after an injury heals

A signal in the brain can predict who will continue to suffer back pain more than a year after an initial injury. This early warning sign could reveal new ways to reverse or prevent pain that lingers long after an injury heals, scientists report online July 1 in Nature Neuroscience.

?We?re very excited about these results,? says study coauthor A. Vania Apkarian of Northwestern University in Evanston, Ill. ?We think they open up a whole new way of looking at chronic pain.?

The study included 39 people with newish back pain, about half of whom still suffered a full year later. These people?s pain had turned chronic, morphing from the pain associated with the original problem to something more devastating. At the start, pain intensity was similar in people with chronic pain and in those who recovered.

But people whose pain turned chronic had an unusually strong connection between two parts of their brains: the nucleus accumbens and the prefrontal cortex. These two regions behaved in tandem, brain scans revealed, so that when one was busy the other was too. The strength of this connection predicted which participants would have lingering pain a full year later: The stronger the connection, the more susceptible a person was to chronic pain.

?This is something we can study,? says neuroscientist Laura Stone of McGill University in Montreal. ?We can figure out how to target this to prevent that transition.?

Earlier studies have catalogued brain differences in people with chronic pain and healthy controls, but researchers never knew whether such differences were the cause of chronic pain or an effect of living with it. This study is the first to uncover a signal that?s present before pain becomes chronic, Stone says.

The study may also link chronic pain development to the brain?s addiction machinery, which includes the nucleus accumbens. ?This is certainly part of the addiction pathway,? Apkarian says. Though the idea hasn?t been tested, he says, chronic pain may stem from the brain essentially becoming addicted to pain.

Stone says the concept of pain co-opting the addiction circuitry in the brain makes a lot of sense, but it?s too early to say whether that idea is right.

Round Town certainly is the epitom? of the All-American experience. There?s at least four baseball fields, a brand new public high school, Round High, a prestigious Catholic college, St. Thomas More Liberal Arts Academy and Seminary, a classic town hall, and a cute little farmer?s market.

But Round Town is hiding a secret. A little ways off from town is a family. A large family. The Lorraines came to town about ten years ago, and instantly fell in love with everything. The Lorraines enjoy Friday night baseball games, send their kids off to Round High, attend various churches throughout the town, participate in local government, and enjoy Saturday mornings at the farmer?s market. They appear to be quite a happy family. But there?s one thing that hasn?t escaped the townspeople: there seems to be more adult-Lorraines than the original count. Though Lorraines have yet to officially open up about their family life, it is well-known the Lorraines are polygamists.

Can the Lorraines prove that it's possible to unite such a large family, despite all their human faults and dynamic relationships? Can they do it before the town tears their family apart? Will their family tear itself apart? Or will the Lorraines be able to successfully convince Round Town that they can be perfectly normal members of the community?

We Still Have One Heart is a literate+ RP devoted to the inner workings of a multi-faceted family in classic America. Characters are based on many interchangeable faceclaims, and role-players are welcome to come and create living, breathing avatars. Reservations last two days and are renewable. This RP will be slow-paced, so it can be juggled with any number of RPs. The GM, although new, is ready to help, entertain and immerse all players, as he draws from years of many different kinds of RP as well as outside life experiences.

Note: This RP is inspired, but not based on, the Sister Wives television series on TLC, which features the Brown family of 1 husband and 4 wives. This family is FLDS, but the religiosity in We Still Have One Heart is fluid and dynamic, and any character of any creed (or lack thereof) is invited.
